Introduction : High motility group (HMG) proteins are transcription factors that are expressed during embryonic development, not in normal adult tissues. The HMGI family consists of 3 proteins, HMGI, HMGI(Y) and HMGA2 (also known as HMGI-C). Experiments with knockout HMGA2 -/- mice yielded a reduced body weight compared to wild-type HMGA2 +/+ mice, which indicates that HMGA2 plays a role in mammalian growth. Interest in HMGA2 has increased recently as it has been found that HMGA2 is expressed in neoplastic tissues and that it apparently has a role in control of cell growth, differentiation and tumorigenesis. HMGA2 is expressed in tumor tissue but not in normal tissue immediately adjacent to the tumor tissue. Studies in peripheral blood show that HMGA2 is not present in normal healthy donors, but is present in the blood of a subset of breast cancer patients. In general, the presence of HMGA2 in the peripheral blood of breast cancer patients has a correlation with poor survival and with a higher histologic grade of the tumor.
